Durham is in south-western Ontario, on Highway 6, two-thirds of the way between Guelph (to the southeast) and Owen Sound (to the north). Highway 6 (north) is intersected by Highway 4 (east/west) in Durham.
From the WINDSOR/SARNIA area, you have a couple options. You can take Highway 21 north, which is a nice drive but a busy road in the summer. You would take #21 to #4, then come east through Walkerton & Hanover, to Durham. This would be the easiest route in terms of navigation.
Another option from Sarnia is to go cross country, north and east to Durham. These roads are generally not as busy, but you’ll have to pay close attention to your map and road signs.
From the GUELPH area, just follow Highway 6 north.
From the TORONTO area, there are many different roads going north from which you can connect to Highway 9. Take Highway 9 west to Arthur and then go north on Highway 6.
From the COLLINGWOOD area go south through the Beaver Valley, till you hit Highway 4. Go west through Flesherton, all the way to Durham. Turn south (left) at the lights in Durham.
